What is the most stable dimension(4x6, 6x6, etc) to use for posts for a large double sided sandblasted sign? Also what is the best way to design the posts so that, say 72''x11.25'', panels can easily be removed/replaced without having remove the top main sign to slide them up/down a mortise?

It sounds like you're describing a series of removable, double-sided tenant panels that will hang under a main permanent panel. If I've got that right, the techniques I use sometimes are double poles with the panel sandwiched between so they can slide out to either side, or angle tabs on each panel that screw to the inside edge of the uprights (only really need them on one side), or even a continous angle or batten up and down the length of the uprights, or finally, that mortise you mention - the mortise can come completely out one side with a removable filler strip to finish it off.
